abstract 1,176,999. The antibiotic bikaverin. CESKOSLOVENSKA AKADEMIE VED. 1 June, 1967 [2 June, 1966], No. 25230/67. Heading C2A. An antibiotic bikaverin, effective against Leishmania brasiliensis, is an acid giving a red colour in acid media and a blue-purple in alkaline media, it is soluble in chloroform, slightly soluble in butanol, benzene, ethyl acetate and vegetable oils; insoluble in water, petroleum ether and hexane; it sublimes above 250‹ C.; m.pt. 315-320‹ C., elementary analysis 59À23% C and 3À07% H; molecular weight 382; empirical formula C 19 H 12 O 9 , ultraviolet absorption as in Fig. 2 (not shown), with maxima at 254 nm. and 278 nm., infra-red absorption as in Fig. 1. The antibiotic is extracted from the mycelium resulting from the production of gibberellins by cultivating Gibberella fujikuroi strain U or Fusarium oxysporum Schlecht strain No. 1-70. The extraction is by means of chloroform, trichloroethylene or a mixture thereof wih methanol or ethanol at pH 1 to 6À5. The broth has preferably been fermented 72 to 96 hours. Purification is obtained by treating the crude antibiotic with petroleum ether or n-hexane to remove impurities, dissolving in chloroform, treating the organic solution with aqueous KOH, filtering off the precipitate, dissolving in chloroform with the aid of hydrochloric acid, washing with water, removing solvent and recrystallizing from chloroform-methanol. A pharmaceutical composition for topical application comprises the antibiotic bikaverin and a carrier.
