titleOfInvention Stable Form of D-(-)-alpha-Aminobenzyl Penicillin and a process for the preparation thereof
abstract 1,160,102. Ampicillin. BRISTOL-MYERS CO. 9 Aug., 1966 [9 Aug., 1965], No. 35668/66. Heading C2A. Form III D-( - ) α-aminobenzylpenicillin is a stable crystalline form of this penicillin, substantially free of water in the chemically bound state, i.e. contains less than about 1À8% water, and has the infra-red absorption spectrum shown in Fig. I. It is prepared by contacting an acid addition salt of D-( - ) α-aminobenzylpenicillin with one or more amines having the formula wherein R is an alkyl group having from 10 to 15 C atoms; R<SP>1</SP> is H or C 1 to C 6 alkyl, and R<SP>2</SP>, R<SP>3</SP> and R<SP>4</SP> are each alkyl groups having an aggregate of from 11 to 14 carbon atoms, in a water-immiscible organic solvent containing at least a minor amount of water, at a temperature of at least 60‹ C. The acid addition salt may be with an organic or inorganic acid, preferably an aryl sulphonic acid having the formula R<SP>5</SP>SO 3 H, wherein R<SP>5</SP> is in which R<SP>6</SP>, R<SP>7</SP>, R<SP>8</SP> and R<SP>9</SP> are hydrogen, C 1 to C 12 alkyl, C 1 to C 6 alkoxy, C 1 to C 6 alkylthio, NO 2 , C 1 to C 4 alkanoylamino, C 1 to C 6 alkanoyloxy, sulphamyl, chloro, iodo, bromo, fluoro, trifluoromethyl, C 1 to C 6 alkylsulphonyl, carbo C 1 to C 6 alkoxy, benzyl, phenethyl, cyclohexyl, cyclohexyl or cyclopentyl. The penicillin is distinguished from the anhydrous ampicillins of Specifications 1,003,868 and 1,016,875. Specifications 873,049, 962,719 and 1,011,843 are also referred to. A pharmaceutical composition comprises the above ampicillin and a carrier for human or veterinary use. It may be added to animal feeds and used for the treatment of mastitis in cattle.
