abstract | 1,153,973. Substituted 1-azaspiro[4,5]decane (and [4,4]nonane) - 1 - propanols; penicillin salts. UPJOHN CO. 10 Feb., 1967 [14 March, 1966], No. 6550/67. Headings C2A and C2C. Novel compounds of the formula wherein γ is H, F, Cl, Br and C 1-4 alkyl, R and R<SP>1</SP> are H or CH 3 and n is 1 or 2 and the acid addition salts thereof are prepared by reacting in an anhydrous solvent system the appropriately substituted phenyl magnesium halide with a C 1-4 alkyl α - methyl - 2R - 3R<SP>1</SP> - 1 - azaspiro- [4,5] - decane (or [4,4]nonane) - 1 - propionate, resulting from the condensation of a 2-R-3- R<SP>1</SP>-1-azaspiro[4,5]decane (or [4,4]nonane) with a C 1-4 alkyl methacrylate, and decomposing the reaction mixture with acidified ice water. Salts of the above novel compounds with penicillins, particularly benzyl penicillin, are prepared by either by the neutralization of the novel free base with the acid form of a penicillin or by a metathetical exchange of the anion of an acid addition salt of a compound of the above general formula with an anionic form of penicillin. These salts have solubility characteristics which makes them useful in the isolation and purification of penicillin. Pharmaceutical compositions, which may be used as sedatives and tranquilizers, contain the above novel compounds or acid addition salts thereof as the active ingredient. |
