abstract 1,153,818. Dihydroguinoline - N - carboxylic and thioncarboxylic esters. BRISTOL-MYERS CO. 10 Aug., 1967 [15 Aug., 1966], No. 36729/ 67. Heading C2C. Novel alkyl esters of 1,4-dihydroguinoline-N- carboxylic- and thioncarboxylic-acids, of formula where R<SP>1</SP> is C 1 -C 6 alkyl and X is oxygen or sulphur, are prepared by the following procedure: (1) hydrogenating an ethylenically unsaturated compound of formula to give the corresponding alkylene derivative, in each case Y being oxygen or sulphur, and the groups R<SP>3</SP> being either separate C 1 -C 6 alkyl groups or together forming a -(CH 2 ) 2 - or -(CH 2 ) 3 - group which constitutes a heterocycle with -Y-CH-Y-; (2) acylating the latter compound with a C 1 -C 6 alkyl chloroformate or chlorothionformate to give a compound of formula and then either (3)(a) hydrolysing this compound to effect ring closure, giving a mixture of compounds of formulĀµ which compounds are then heated, removing the -Y-H or -Y-alkyl group respectively, either individually after separation, or as a mixture and giving the required product; or (3)(b) simultaneously hydrolysing and removing the said group from the compounds to provide the required product in a single step. 2 - Hydroxy - 1 - carbethoxy - 1,2,3,4 - tetrahydroguinoline is esterified to the corresponding 2-ethoxy compound with HCl and ethanol. Pharmaceutical compositions having tranquillizing activity, comprise the inventive compound with an orally or parenterally acceptable carrier; they may take the form of tablets, powders, capsules, granules, suspensions or solutions; the invention includes a method for tranquillizing animals by administering the active agent or its compositions.
