The invention comprises (a) compounds of formula wherein A is C 1-8 alkylene, X is hydrogen, C 1-8 alkyl, C 1-8 alkoxy, phenoxy, hydroxy, alkylthio, nitro, amino, dialkylamino, halo or trifluoromethyl, Y is -CH 2 -, -CH 2 CH 2 -, -O- or -S-, R is C 1-8 alkyl, aryl, aromatic heterocyclyl, cycloalkyl, aralkyl, aromatic heterocyclyl-alkyl, cycloalkyl-alkyl, allyl or propargyl, R<SP>1</SP> is hydrogen or C 1-8 alkyl, R<SP>2</SP> is hydrogen, hydroxy, acyloxy or C 1-10 alkoxy, R<SP>3</SP> is aryl or aromatic heterocyclyl, n is zero or one, and NB is amino, C 1-8 alkylamino, di (C 1-8 alkylamino), phenyl (C 1-8 alkyl) amino, N-C 1-8 alkyl-N-phenyl-(C 1-8 alkyl)-amino, pyridyl, piperidino, mono or di-C 1-8 alkyl-piperidino, C 1-8 alkoxypiperidino, 2-, 3- or 4-piperidyl or N-C 1-8 alkyl, piperidyl, pyrrolidino, mono or di-C 1-8 alkyl-pyrrolidino,C 1-8 alkoxy-pyrrolidino, C 1-8 alkoxy-pyrrolidino, 2- or 3-pyrrolidyl or N-C 1-8 alkyl-pyrrolidyl, morpholino, mono- or di-C 1-8 alkyl-morpholino, C 1-8 alkoxy-morpholino, thiamorpholino, mono- or di-C 1-8 alkoxy-thiamorpholino, piperazino, mono or di- C 1-8 alkyl piperazino, C 1-8 alkoxy-piperazino, N<SP>1</SP>-aryl, aralkyl, aromatic heterocyclyl or aromatic heterocyclyl-alkyl-piperazino or X- substituted-cinnamyl; and (b) N,N-dimethyl-3- (2 - methyl - 2 - phenyl - 1 - indonylidene) propylamine hydrochloride, stereo-isomers, acid addition salts and quaternary ammonium salts thereof, and their preparation, by reaction of compounds of formula with a Grignard reagent Hal-Mg-A-NB to give a compound of the invention wherein R<SP>2</SP> is hydroxy followed if desired by (1) etherification, (ii) esterification or (iii) dehydration to give compound (b) above or an analogue thereof, followed by hydrogenation of the double bond. The compounds of the invention have analgesic activity. 2-benzyl-2-phenyl-1-indanone is prepared by reaction of 2-phenyl-1-indanone with sodium hydride and benzyl chloride. |
