titleOfInvention Phenyl benzoxazocineones and their preparation
abstract 1,148,718. Substituted benz-(f)-2,5-oxazocine- 4-ones. REXALL DRUG & CHEMICAL CO. 10 May, 1966 [10 May, 1965], No. 1931/68. Divided out of 1,148,717. Heading C2C. Novel 5-C 1-6 -alkyl-1-phenyl benz-(f)-2,5-oxazocine-4-one having the formula where R is C 1-6 alkyl, and the benzenoid portion of the fused ring structure and/or the phenyl group may bear one or more C 1-6 alkyl, C 1-6 alkoxy, halo or CF 3 substitutents, are obtained by subjecting a 2-(N-lower alkylaminomethyl)- benzhydrol of the general formula where R is C 1-6 alkyl and either or both of the benzene rings may bear one or more C 1-6 alkyl, C 1-6 alkoxy, halo or CF 3 substituents, to a threestage reaction sequence involving, (a) treatment with haloacetyl halide, (b) treatment of the resulting product with a base, and (c) treatment of the product of (b) with potassium-t-butoxide in the presence of an organic solvent such as dimethyl sulphoxide. In an example, 2-(N- methylaminomethyl)-benzhydrol is reacted with chloracetyl chloride to form 2-(N-methylchloroacetamidomethyl)-benzhydrol chloroacetate which is treated with methanolic NaOH to give 2-(N- methylchloroacetamido methyl)-benzhydrol. The latter is treated with potassium t-butoxide in dimethyl sulphoxide to give 5-methyl-1-phenyl- 1,3,4,6 - tetrahydro - 5H - benz - ( f ) - 2,5 - oxazocane-4-one.
