titleOfInvention Improvements in or relating to novel steroids and the manufacture thereof
abstract 1,145,063. Steroids. UPJOHN CO. 6 April, 1966 [7 April, 1965], No. 15373/66. Heading C2U. Novel compounds of the formula wherein S is a generic expression denoting α- and #-double bonds and mixtures thereof; the 4(5) carbon linkage is a single or a double bond with the proviso that the 5-hydrogen atom is absent when the 4(5)-carbon atom linkage is a double bond, R, R 1 and R 2 are selected from hydrogen and methyl; Y is Z being hydrogen, tetrahydropyranyl or the acyl radical of a (C 1 -C 12 ) hydrocarbon carboxylic acid, are obtained by heating a corresponding compound of the formula with an acid. Starting 3-keto-#<SP>4</SP>-androstenes may be saturated by hydrogenation and further reduced to form a mixture of 3-(α and #)- hydroxy-androstanes. A 17-acylate grouping if present may be hydrolysed to form a 17- hydroxy compound. 3-Desoxy starting compounds are obtained by converting the 3- oxygenated compounds in the corresponding thioketal by reaction with an alkanethiol, followed by desulphurization of the latter, e.g. with sodium in liquid ammonia. The 3-dihydropyranyl ether starting compounds may be obtained by reacting a 3-17-dihydroxy-5α- androstane 17-acylate with dihydropyran in the presence of a Lewis acid, and if desired the 17-acylate is then hydrolysed. Pharmaceutical compositions having anabolic, anti-fertility, anti-androgenic, anti-estrogenic, CNS-regulating, antimicrobial activities and also useful in the prevention of atherosclerosis and thromboembolic disease and containing the above compounds are administered orally or parenterally.
