abstract | 1,144,215. Preparing erythrocytes for immunochemical use. ORGANON LABORATORIES Ltd. 28 June, 1966 [28 July, 1965], No. 28963/66. Heading B1X. An aqueous suspension of erythrocytes treated with one or more tanning agents, is dried and heated at above 30‹C, to provide a product which can be compressed into tablets for storage and use in immunochemical determination. The erythrocytes may first be sensitized with an antigen or this may be done subsequently. Other materials may be present in the suspension, e.g. tableting auxiliaries such as fillers (saccharose, mannitol, lactose, urea), lubricants (boric acid, starch, carbowax (Trade Mark) and magnesium stearate), disintegration agents (starch, alginic acid) and substances used in the determination e.g. buffers (monopotassium and disodium phosphate), sodium chloride, EDTA and albumin. Alternatively, these auxiliaries may be mixed individually or as a spray-dried mixture, with the treated ethythrocytes before tableting. The drying may be performed by lyophilisation, and the heating by fluidisation in a dry gas stream at 50- 120‹C for 1-10 mins, but preferably drying and heating are performed simultaneously by spray drying e.g. at 75-140‹C. In the Examples the erythrocytes were tanned with formalin with or without tannin, but the use of inulin, hydroquinine, bis-diazo benzidine, acetaldehyde, pyruvic acid aldehyde and glyoxal as tanning agents (mordants) is mentioned. In the Examples, the erythrocytes are sensitized with HCG, but serum gonadotrophin, growth hormone, serium albumins, gamma globulins, insulin, luteinizing hormone and tetanus toxoid are other antigens mentioned. |
