titleOfInvention | Novel imidazole derivatives and a process for the manufacture thereof |
abstract | 1,139,455. Imidazole derivatives. F. HOFFMANN-LA ROCHE & CO. A.G. 9 June, 1967 [10 June, 1966], No. 26766/67. Heading C2C. Novel compounds of the general formula wherein R<SP>1</SP> and R<SP>2</SP> each represent an optional C 1-7 alkyl group and Z represents a grouping of formula or, when at least one of R<SP>1</SP> and R<SP>2</SP> is present, a grouping of the formula or, when R<SP>1</SP> and R<SP>2</SP> are both absent, a grouping of the formula and pharmaceutically acceptable salts thereof: in which groupings R<SP>3</SP> represents a halogen atom or a hydroxyl, lower-alkoxy, lower-alkenyloxy, lower-alkynyloxy, halo - (lower-alkoxy), polyhalo-(lower-alkoxy) or halo-(lower-alkynyloxy) group or a phenoxy group which may carry one or more halo, lower-alkyl or lower-alkoxy substituents, Ac represents a lower-alkanoyl group or a benzoyl group which may carry one or more halo, lower-alkyl or lower-alkoxy substituents, R<SP>4</SP> represents a lower-alkanoyl group, R<SP>30</SP> represents a lower-alkenyloxy, lower-alkynyloxy, halo-(lower-alkoxy), polyhalo-(lower-alkoxy) or halo-(lower-alkynyloxy) group or a phenoxy group which may carry one or more halo, lower-alkyl or lower-alkoxy substituents and Ac<SP>1</SP> represents a benzoyl group which may carry one or more halo, lower-alkyl or loweralkoxy substituents, are prepared by reacting a 2-nitro-imidazol with a compound of the general formula and, if desired, converting the resulting alcohol into its alkanoic acid ester, or by reacting a compound of the general formula wherein of R<SP>1</SP> and R<SP>2</SP> at least one is present with a compound of the formula to form an alcohol of the above formula and, if desired, acylating the said alcohol to give an ester of the above formula or oxidizing the said alcohol formed with a chromate solution to form a ketone of the above formula or treating an alcohol in which R 3 is present and represents the halogen with a base to form a compound containing the epoxide grouping, or by reacting an imidazole of the last general formula above with a halo-acetone to form a ketone of the first general formula above, or reacting a ketone with semicarbazide or a salt thereof to form a semicarbazone of the first formula above and, in any case, converting, if desired, the product into a pharmaceutically acceptable salt thereof. 1,2 - Epoxy - 3 - [3 - iodo - propyn - (2) - yloxy]. propane is prepared from 3-iodopropargyl alcohol and epichlorohydrin. Pharmaceutical compositions having antimicrobial activity and in conventional form for oral, rectal, parenteral or topical application, comprise an above novel compound and one or more carriers or diluents therefor. |
