titleOfInvention Cephalosporanic acid derivatives
abstract 1,138,768. 7 - (1,2,5 - Thiadiazole - 3 - acetamido) - cephalosporanic acids. BRISTOLMYERS CO. 21 July, 1966 [21 July, 1965], No. 32792/66. Heading C2A. The invention relates to 7-(1,2,5-thiadiazole- 3-acetamido)-cephalosporanic acids of Formula I wherein A is H, OH, C 2 to C 8 alkanoyloxy, benzoyloxy, quaternary ammonium or when taken with M is a C-O bond; and M is H, a pharmaceutically acceptable non-toxic cation, an anionic charge when A is a quaternary NH 4 radical, or when taken with M is a monovalent C-O bond. Typical compounds are 7-(1,2,5- thiadiazole-3-acetamido)-cephalosporanic acid, 3 - pyridinium-methyl - 7 - (1,2,5 - thiadiazole - 3-acetamido)-decephalosporanic acid inner salt, 3 - hydroxymethyl - 7 - (1,2,5 - thiadiazole - 3 - acetamido) - decephalosporanic acid and 3 - methyl - 7 - (1,2,5 - thiadiazole - 3 - acetamido) - decephalosporanic acid. The compounds are prepared by acylating a compound of formula with an acid or functional derivative thereof of formula wherein A has the above meaning. When A is alkanoyloxy in the resulting compound, further reaction with a tertiary amine, e.g. pyridine, picoline or lutidine produces the corresponding quaternary ammonium inner salt. The salts may be inorganic, e.g. sodium or organic, e.g. procaine. Therapeutic compositions for administration to man and animals comprises an antibacterially active 7 - (1,2,5 - thiadiazole - 3 - acetamido) - cephalosporanic acid of Formula I and a carrier. The compositions may be used for treating mastitis in cattle.
