abstract | 1,136,347. Recovery of basic antibiotics. CHAS. PFIZER & CO. Inc. 8 Feb., 1967, No. 6119/67. Heading C2A. [Also in Division B1] The adsorption of a basic antibiotic upon a particulate carboxylic acid type cation-exchange resin is obtained by flowing an impure solution of said antibiotic, e.g. whole or clarified fermentation broth, in a substantially horizontal direction through a free suspension of said resin until the adsorption of the antibiotic content of said solution by said resin substantially ceases and then separating the antibiotic-depleted solution from the resin adsorbate, said resin being present in a proportion of about 0À1 to about 0À8 volume per volume of solution in the vessel confining said resin. The resin should have a particle size of about 100 mesh or coarser (U.S. Standard Sieve) and a specific gravity of about 1À0. Preferably, the resin is used in the form of a salt with a monovalent cation, e.g. sodium or ammonium salt. Copolymers of acrylic acid or methacrylic acid with about 2 to 8 per cent of divinylbenzene are preferred resins. Specified antibiotics are streptomycin. neomycin, viomycin, polymycin, dihydrostreptomycin, hydroxystreptomycin, streptothricin and mannosidostreptomycin. For streptomycin, the pH of the broth is adjusted to pH 7 to 8. The adsorption is preferably carried out in two or more troughs 12 connected in series and each fitted with a concentric shaft 20 carrying a plurality of vanes 24 one of which carries a rubber wiper 26 for preventing agglomeration of the resin in the lower part of the trough. The flow of antibiotic is such that the effluent from the last vessel is substantially free of antibiotic content. After washing with water, the antibiotic - saturated resin is eluted with 2N.H 2 SO 4 , neutralized and the streptomycin recovered therefrom. Alternatively, rotating drum apparatus may be used. The resin may be maintained in the suspended state by mechanical, ultrasonic or gas bubble means. |
