titleOfInvention Process for the production of basically substituted arylalkyl ketones
abstract Novel basically substituted arylalkyl ketones of the general formula <FORM:1112096/C2/1> wherein X and Y independently of each other represent oxygen or sulphur; R1 represents a divalent hydrocarbon radical having 2 to 11 carbon atoms, which links X and Y by way of 2 to 4 carbon atoms and has no unsaturated bonds at the two terminal carbon atoms; R2, R3 and R4 independently of each other represent hydrogen, low alkyl or alkoxy radicals or chlorine atoms, and when R4 is hydrogen, R2 and R3 also represent the methylenedioxy radical; and A represents a straight or branched chain alkylidene or alkylene radical having at most 4 carbon atoms, and acid addition salts thereof may be prepared by reacting an azaspiro compound of the general formula <FORM:1112096/C2/2> with a reactive ester of an arylalkyl hydroxy ketone of the general formula <FORM:1112096/C2/3> by the Mannich reaction of formaldehyde, a mineral acid salt of a compound of the second general formula and an arylalkyl ketone of the formula <FORM:1112096/C2/4> wherein R5 represents hydrogen, the methyl or ethyl radical; or by reaction of an azaspiro compound of the second general formula with an unsaturated ketone of the formula <FORM:1112096/C2/5> wherein R6, R7 and R8 represent hydrogen o alkyl radicals, followed in each method by optional conversion to a corresponding acid addition salt. Pharmaceutical preparations comprising as active ingredient at least one basically substituted arylalkyl ketone of the first general formula or a pharmaceutically acceptable acid addition salt thereof, together with at least one pharmaceutical carrier, have useful sedative, muscle relaxing, analgesic and appetite inhibitory action. Azaspiro compounds of the second general formula, such as 1,4-dioxa-8-azaspiro[4,5]decane and 1,5-dioxa-9-azaspiro[5,5]undecane may be prepared from 4,4-dihydroxypiperidine hydrochloride and the required dihydroxy-, hydroxy-, dimercapto- or mercapto-compounds.
