abstract An aqueous solution of flavin-adenine dinucleotide (F.A.D.), e.g. from cell extracts or fermentation broths, is purified by adsorbing the F.A.D. on to a phenolic cation-exchange resin at a pH of about 1.0-3.0 and then eluting the F.A.D. with an aqueous eluant having a pH of about 3.0-10.0. The adsorbed F.A.D. may be washed with an acidic aqueous solution of pH less than about 3.0 which may contain an inorganic salt such as NaCl or NH4Cl, e.g. a solution of HCl/NaCl. The eluant may comprise water or deionized water or alkaline or acid solutions such a HCl or CH3COOH or buffer solutions having a suitable pH. An aqueous organic eluant containing a water-miscible solvent such as MeOH, EtOH, Me2CO or pyridine may also be used. The cation exchange resins used include those having sulphonic acid and methyl sulphonic acid radicals and may be used in the free or salt form, preferably the sodium or ammonium salt form. The crude F.A.D. solution preferably contins an inorganic salt such as NaCl or NH4Cl in a concentration of about 1 mol. per litre. The cation exchange resin is preferably used in a column and F.A.D. may be recovered from the eluate by concentrating by evaporation and adding a suitable organic solvent such as ethanol.
