abstract | Pure, highly viscous, sterilizable hyaluronic acid free from proteins, antigens and pyrogens is prepared by producing by precipitation, a dry powder from animal organs containing hyaluronic acid, denaturing the protein fraction of the dry powder by heating it in aqueous suspension above pH 7.0, decomposing said protein fraction by means of proteolytic ferments and removing the resulting free amino acids and mineral salts by treatment with ion exchangers, the resulting impure hyaluronic acid being then adjusted to pH 3-4 whereby the remaining impurities form with a portion of the hyaluronic acid an insuluble complex which is separated by high speed centrifuging. The initial dry powder may be obtained by precipitation with acetone, and the protein fraction decomposed by a hydrolase mixture of aspergillus orizae. The product has application as a substitute for the vitreous body of the eye (see Division A5).ALSO:Hyaluronic acid which is pure, highly viscous, sterilizable and is free from proteins, antigens and pyrogens may be used in aqueous solution as a substitute for the vitreous body in the human eye. The solution is prepared from animal organs containing hyaluronic acid (see Div. C3). |
