titleOfInvention | 17ª-halo-alkenyl and alkynyl steroids |
abstract | Novel steroids of the formulae <FORM:1069802/C2/1> (wherein Y is a two or three carbon unsaturated halogenated hydrocarbon group, and R1 is H or acetyl) are prepared by reacting the appropriate D 4,9(10)-3-ones with lithium aluminium hydride tri-tertiary butoxide in an inert organic solvent to produce the corresponding D 4,9(10)-3b -ols and if desired, acetylating these to give the 3b -acetoxy compounds, or by acetylating the 17b -hydroxy-D 4,9(10)-3-ones to produce the D 3,5(10) 9(11)-3-acetoxy compounds (and concomitantly the 17-acetates of the starting materials). 17a - Y - 17b - Hydroxy - estra - 4,9(10)-dien-3-ones are prepared from 3-methoxy-estra-2,5(10)-dien-17-one and halo-alkynes or -alkenes or their metal derivatives with subsequent hydrolysis to 5(10)-en-3-ones and reaction of these with bromine in pyridine or with a perbromide hydrobromide. 17a -(31,31,31 -Trifluoropropenyl)-derivatives are prepared by reduction of the trifluoropropynyl compounds. 17b - Acetoxy - 17a - Y - estra - 4,9(10) - dien-3-ones are prepared by acetylation of the 17b -ols. The steroids of the invention, which are stated to be progestational agents, may be made up into pharmaceutical compositions for oral or parenteral administration with suitable carriers. |
