titleOfInvention | Improvements in or relating to 19-amino-10-a-androstane compounds |
abstract | 1,061,026. 19-Amino-10α-androstane compounds. SYNTEX CORPORATION. Nov. 8, 1963 [Dec. 20, 1962], No. 44139/63. Heading C2U. Novel steroids of the formula (wherein R is hydrogen or C 1-4 alkyl; R<SP>1</SP> is hydrogen or hydrocarbon carboxylic acyl of less than 12 carbon atoms which is optionally substituted; and R<SP>2</SP> is hydrogen, C 1-8 alkyl, C 2-8 alkenyl or C 2-8 alkynyl; provided that when R is hydrogen and R<SP>2</SP> is not hydrogen, R<SP>1</SP> is hydrogen) are prepared by converting 19- hydroxy - 10α - testosterone to the 19 - trityl ether, acylating and treating with acid to remove the trityl group, to give, for example, 19- hydroxy - 10α - testosterone 17 - acetate, oxidizing this to the 19-oic acid, treating this with oxalyl chloride to give the acid chloride, treating this with ammonia or the amine NHRR to give the amide, reducing this with lithium aluminium hydride to give the corresponding 19 - amino - #<SP>4</SP> - 10α - androstene- 3#,17# - diol, oxidizing this with 2,3 - dichloro - 5,6 - dicyano - 1,4 - benzoquinone to give the 3-one and, if required, converting this to the 3-ketal, oxidizing to the 17-one, converting to the 17#-hydroxy-17-α-hydrocarbyl compounds and hydrolysing the ketal group. The products may be acylated. The steroids of the invention, which are stated to be anabolic-androgenic agents and to have anti-estrogenic, anti-gonadotrophic, antifibrillatory, appetite-stimulating, antipyretic and analgesic properties, may be made up into pharmaceutical compositions with suitable carriers. |
