titleOfInvention Occult blood compositions using quinoline compounds for increased sensitivity
abstract Compositions for detecting occult blood in body fluids comprise an indicator the oxidation of which is catalysed by the prosthetic group of haemoglobin, an oxidising agent for the indicator and a potentiating agent of the formula:- <FORM:1057056/B1-B2/1> where R1 is H, CH3 or OH; R2 is H, CH3 or thiophenyl; R3 is H, CH3 or:- <FORM:1057056/B1-B2/2> R4 is H, CH3 or 0CH3; R5 is H or CH3; and R6 is H, CH3, OH or NH2. Specified indicators are o-tolidine, o-toluidine, p-toluidine, o-phenylenediamine, N:N1-dimethyl-p-phenylenediamine, p-anisidine, dianisidine, o-, m-, and p-cresols, a - and b -naphthols, catechol, guaiacol and pyrogallol. Specified oxidizing agents are inorganic and organic peroxides, namely hydroperoxide of cumene, p-menthane diisoproplybenzene and 2:5-dimethyl hexane. Specified potentiating agents include quinoline itself, quinine, cincholine and a number of other substituted quinolines. In the composition, the organic hydroperoxide is preferably in the form of encapsulated particles prepared in dispersion using gelatin, algin, carrageenin, casein or albumin and hardened by treatment with a dialdehyde polysaccharide as described in Specification 981,955. The procedure is described in detail. Other ingredients may comprise buffering systems (citrate, tartrate, phthalate, acetate, phosphate) giving a pH of 4-7 emulsifiers for the hydroperoxide (acacia mucilage, gum arabic, polyvinyl alcohol, carboxyvinyl polymer), surface active agents (sodium lauryl sulphate, dioctyl sodium sulphosuccinate, polyoxyethylenenonylphenol) which improve the evenness of application to carrier materials and solvents (chloroform, ethylene dichloride, ethyl acetate and benzene) for the indicator and potentiator. The compositions may be in the form of tablets, a mixture to be added to water, or impregnated on to bibulous materials e.g. paper or wood.
