abstract | The invention comprises compounds of the formula <FORM:1047110/C2/1> wherein R represents a C1- 7 alkyl or phenyl group, S1 represents one or two substituents selected from halogen, C1- 7 alkyl, C1- 7 alkoxy, hydroxy, nitro and acetamido, which substituents may differ from each other, and the broken lines indicate that the substituent atom or groups attached thereby are optional, and acid addition salts thereof; and a process for their preparation comprising reacting an acetophenone of the formula <FORM:1047110/C2/2> with formaldehyde and an oxazine of the formula <FORM:1047110/C2/3> or an acid addition salt thereof, and, if desired, converting the reaction product into an acid addition salt or the free base as the case may require. 6,6 - Diphenyl - 3,4,5,6 - tetrahydro - 2H - 1,3 - oxazine hydrochloride is prepared by reacting 3-benzylamino-propiophenone hydrochloride with phenylmagnesium bromide to produce a -phenyl-a - (2 - benzylaminoethyl)benzyl alcohol, debenzylating this compound by hydrogenolysis to form a - phenyl - a - (2 - aminoethyl)benzyl alcohol, reacting this compound with formaldehyde and treating the reaction product with hydrogen chloride. Pharmaceutical compositions having hypotensive activity, for oral or parenteral administration, comprise a compound of the invention together with a pharmaceutical carrier. |
