titleOfInvention Novel d-nor steroids and processes for their manufacture
abstract The invention comprises D-nor-steroids and a process for the production thereof by dissolving a steroid substituted in the D-ring by a keto group and by a diazo group in a position a - to the keto group, in an inert solvent containing a compound which can be added across the double bond of a ketene, photolysing the solution and isolating the D-nor-steroid thus produced. By employing a 16-diazo-17-keto androstane or estrane dissolved in an inert solvent containing water, ammonia, an alcohol or primary or sec. amine, there is produced a D-nor-16b -carboxylic acid, carboxylic amide, carboxylic acid ester or -N-mono- or disubstituted-carboxylic acid amide. Further transformations may be effected, e.g. by converting the D-nor-16b -carboxylic acid to the corresponding acid chloride, followed by reaction with an organo-metallic compound or with diazomethane to form a D-nor-pregnane derivative, degrading the D-nor-pregnane derivative to obtain a D-nor-androstane derivative which on pyrolysis gives a D-nor-estrane derivative. The starting a -diazo-keto steroid may be obtained by reacting the corresponding a -oximino-keto steroid with chloramine. Pharmaceutical compositions containing the above compounds and having estrogenic, androgenic, anti-inflammatory, cortical, cholesterol-lowering, anabolic, progestational and antifertility activity are administered orally or parenterally.
