abstract | The present invention relates to a method for assaying nucleic acids of the DNA type, capable of being salted out in a flow liquid of an affinity chromatography column, containing a support on which said DNA aptamers are immobilized, said nucleic acids DNA type representing the entire DNA aptamers, or a fragment thereof, said method comprising the steps of: a) providing a sample of said flow liquid likely to contain DNA nucleic acids released from the support of the column affinity chromatography; b) performing PCR amplification of the DNA nucleic acids contained in said sample, using a reaction mixture comprising (i) a pair of nucleotide non-degenerate sense and antisense primers specific for said DNA nucleic acids and (ii) ) a fluorescent intercalating agent, to obtain an amplification product in the form of double-stranded DNA nucleic acids complexed with said fluorescent intercalating agent; and c) quantifying the DNA aptamers, or fragments thereof, by measuring the level of a fluorescence signal generated by said amplification product and comparing this level of fluorescence to that obtained by a standard range generated by a known concentration of said aptamer DNA. |
