abstract | The invention relates to a method for inducing targeted meiotic recombination in a eukaryotic host cell, by co-expressing in said cell a first ME chimeric protein comprising an effector domain DE capable of inducing the formation of double-strand breaks. strand in the DNA during meiotic recombination and a DI1 interaction domain consisting of the first partner of a protein-protein selective affinity couple, and a second chimeric protein comprising a DI2 interaction domain consisting of the second partner of said protein-protein selective affinity pair and a DC targeting domain capable of selectively binding to a target sequence present in the DNA of said host cell. The protein-protein interaction between the DI1 and DI2 domains allows the formation of a non-covalent complex between the ME module and the MC module, and the induction of double-strand breaks by the DE effector domain near the targeted sequence by the DC targeting domain. |
