abstract | The present invention relates to novel microparticles of amphiphilic polyamino acids carriers of active principle (s) -PA-, in particular protein (s) and peptide (s), as well as new modified-release pharmaceutical formulations containing said microparticles PA The object of the invention is to develop new PA-loaded microparticles obtained by aggregation of amphiphilic polyamino acid nanoparticles, and having improved properties, especially in dry solid form, with regard to their dispersibility, and with regard to the reconstituted suspension, its stability and its ability to be easily handled and injected. The invention relates in a first aspect, microparticles of amphiphilic polyamino acid (PO) containing at least one PA (non-covalently associated ), spontaneously forming a colloidal suspension of nanoparticles in water, at pH 7.0, in isotonic condition; said microparticles being characterized. in that they are obtained by atomization of a solution or a colloidal suspension of PO comprising at least one PA, b. by cutting between 0.5 and 100 microns, c. and in that they are dispersible in colloidal suspension.The invention also relates to the process for preparing these microparticles, a liquid formulation comprising a suspension of these PO / PA microparticles, a method and a kit for reconstituting this formulation, and a dry form of this formulation. |
