abstract A method for quantitatively evaluating the overall and specific DNA repair capabilities of a biological medium, evaluating the excision / resynthesis capabilities of said medium and its applications.The method comprises the steps of: (a) preparing a range of plasmids each having distinct lesions of the DNA, (b) the characterization of the lesions present on each of the plasmids of said range of plasmids, (c) the deposition of the different plasmids of said range of plasmids and of at least one super-wound plasmid controls without lesions on a single solid support, according to a pre-established scheme A, to form a functionalized support divided into distinct zones A1 to Ax, x corresponding to an integer equal to the number of biological media to be tested simultaneously, each zone A1 to Ax comprising said range of plasmids, (d) incubating said functionalized support obtained in step (c) with different solutions of repairing, (e) at least one washing of said functionalized support, (f) direct or indirect measurement of the signal produced by the marker incorporated in the DNA during the repair reaction of step (d), (g) recording and quantifying the signal corresponding to each plasmid deposition and (h) determining the ratio of the signals of the plasmids comprising the lesions to the control plasmid deposited jointly.
