titleOfInvention METHOD FOR SCREENING BIOMOLECULE BANKS AND APPLICATIONS THEREOF
abstract Method for screening, without amplification, of radioactive biomolecule banks, products of the obtained biomolecule banks as well as application of the method to the identification of molecules capable of selectively binding to a particular tissue or organ, useful for the development of new therapeutic compounds, contrast agents for medical imaging and drug targeting. The method for screening a library of biomolecules is characterized in that it comprises at least the following steps: (1) the administration, to at least one animal, of a library of biomolecules, of the following general formula: propyl -3H-NH-Yaa (R, S) Phe (PO2- CH2) (R, S) Leu-Yaa'-NH2, in which Yaa and Yaa 'represent positions in which the 20 natural amino acids appear and of which each biomolecule is marked beforehand with a suitable radioactive isotope, (2) the sacrifice of at least one of the animals and the analysis of the tissue distribution of the radioactivity of the administered biomolecules, from sections of tissues or organs removed , using suitable imagers, (3) the selection of tissue or organ sections in which a radioactivity signal is detected, (4) isolation, from said sections tissues or organs, selected in step (3) of radioactive fractions, by extraction techniques and / or chromatography and (5) characterization from the radioactive fractions obtained in step (4) of said biomolecule (s), by suitable analysis techniques, such as chromatography and / or mass spectrometry. As a variant, when possible, step (2) comprises the analysis of the tissue distribution of the radioactivity of the administered biomolecules, using suitable imagers, in vitro, from a biological sample previously extracted and selected from the group consisting of cells and sections of tissues or organs. This variant therefore does not require the sacrifice of the animal.
