abstract The invention relates to a method for circularizing an oligonucleotide comprising: - the incubation of a double-stranded nucleic acid comprising a target sequence, with. either a single-stranded oligonucleotide a) comprising a central part capable of binding to said target sequence, linked by spacer sequences to 5 'and 3' terminal sequences, themselves complementary to the sequence of an oligonucleotide matrix,. either a single-stranded oligonucleotide b) whose terminal sequences in 5 ′ and 3 ′ can be fixed adjacent to the target sequence, the incubation step being carried out under conditions allowing the fixation of the single-stranded oligonucleotide a) or b) at the target sequence by formation of reversible bonds, which leads to the winding of the oligonucleotide around its target, - in the first case a), bringing the complex formed into contact with said oligonucleotide matrix , under conditions allowing the hybridization of the 5 ′ and 3 ′ ends, adjacent to the matrix, - the ligation of the hybridized ends, so as to circularize the oligonucleotide around the double-stranded nucleic acid and, if one so wishes, - isolation of the circularized structure, where the target nucleic acid is padlocked with the circular oligonucleotide, followed where appropriate by its purification. Application of the process and the structures obtained in diagnosis and therapy.
