abstract | New farnesyl transferase inhibitors of general formula (I), their preparation and the pharmaceutical compositions which contain them. (CF DRAWING IN BOPI) In general formula (I), R1 represents YS-A1 - (Y = hydrogen atom, amino acid residue, fatty acid residue, alkyl or alkoxycarbonyl radical or an R4 -S- radical in which R4 represents an alkyl radical containing 1 to 4 carbon atoms optionally substituted by a phenyl radical or a radical of general formula: (CF DRAWING IN BOPI) in which A1, X, X1, Y1, R'1, R2, R '2 and R are defined as below, and A1 = alkylene radical containing 1 to 4 carbon atoms optionally substituted in alpha of the group> C (X1) (Y1) by an amino or alkylamino, dialkylamio, alkanoylamino or alkoxycarbonylamino radical) , X1 and Y1 each represent a hydrogen atom or together with the carbon atom to which they are attached form a group> C = O, R'1 represents hydrogen or an alkyl radical containing 1 to 6 carbon atoms, X represents an oxygen or sulfur atom, R2 represents an alkyl, alkenyl or alkynyl radical containing 1 to 6 a carbon atoms optionally substituted by hydroxy, alkoxy, mercapto, alkylthio, alkylsulfinyl or alkylsulfonyl, it being understood that, when R2 represents an alkyl radical substituted by a hydroxy radical, R2 can form, with the carboxy radical in alpha, a lactone, R'2 represents hydrogen or an alkyl radical containing 1 to 6 carbon atoms, and R represents a hydrogen atom or an optionally substituted alkyl radical or an optionally substituted phenyl radical, it being understood that the radical (CF DRAWING IN BOPI) is in position -3 or -4 of the naphthyl ring. These new products exhibit anticancer properties. |
