IMMORTALIZED LINES OF CELLS FROM HUMAN ADIPOSE TISSUE, PROCESS FOR THEIR PREPARATION AND THEIR APPLICATIONS |
abstract |
Immortalized lines of cells derived from human adipose tissue, their method of preparation and their applications as a model for the study of the pathophysiology of metabolism and in particular of obesity and diabetes, as tools for the development of drugs for the treatment of pathological conditions related to metabolic disorders such as obesity and diabetes and as drugs. Said lines are formed by preadipocytes containing a nucleic acid fragment comprising at least one immortalizing fragment of a viral oncogene and at least one promoter selected from the group which comprises the promoter of said viral oncogene and a fragment of the regulatory regions of the human gene. vimentin; express at least one of the following proteins: beta1 and beta2-adrenergic receptors, uncoupling protein (UCP), glucose transporters, Glut1 and Glut4, lipoprotein lipase (LPL) and are able to be converted into mature adipocytes, which produce fat and also express alpha2A and beta3 adrenergic receptors and exhibit an inverted Glut4 / Glut1 ratio, relative to said immortalized preadipocytes. |
