abstract | The present invention relates to a process for the enzymatic amplification in vitro of a DNA fragment in which two primers are used, respectively upstream and downstream, of which at least one is a so-called "stepped" primer which consists of a mixture of oligonucleotides, the oligonucleotides constituting the mixture being in equimolar amounts, of different sizes, of sequences complementary to the same region upstream or downstream of said fragment to be amplified and each oligonucleotide having, from its 5 'end, in the 5 direction '-> 3', the same sequence as the smaller oligonucleotides in the mixture. A subject of the present invention is also a diagnostic method and a diagnostic kit, in particular for viral diseases. |
