abstract The invention relates to esters of 5 'deoxyribonucleotides triphosphates (dNTP) or 5' ribonucleotides triphosphates (NTP), characterized in that: a) they carry a group allowing their detection, b) they are substrates for DNA polymerases or RNA polymerases in a process of elongation of nucleotide chains, on a DNA or RNA matrix, c) their integration leads to an end to the elongation of said chain, d) they are hydrolyzable by chemical or enzymatic means, without altering said chain already formed, allowing the restoration of the hydroxyl function on the sugar, by allowing the elongation of the chain from step b) above. The use of these modified nucleotides allows the implementation of a method of sequencing nucleic acids without gel.
