abstract | To obtain, from beta casein, a partial hydrolyzate enriched, on the one hand, in phosphopeptide or peptide fraction 1-25, and, on the other hand, in precursors of beta-casomorphin (these precursors being fragments covering zone 33-97 of the beta casein molecule), by hydrolysis of beta casein by a proteolytic enzyme, this hydrolysis is carried out in a saline medium, causing the appearance in the medium of a precipitate which is separated from the soluble phase, which constitutes the desired partial hydrolyzate. Preferably, this hydrolysis is carried out in a medium with an ionic strength equivalent to that of a solution of monovalent salt of between 0.5 and 4M. To obtain the peptide fraction 1-25 and the precursors of beta-casomorphin, ion exchange chromatography is carried out on this soluble phase, after having demineralized it by electrodialysis. To obtain the peptide fraction 177-183 with antihypertensive and immunostimulating activity, the precipitate obtained is resolubilized, the resulting solution is brought to a pH of 6-9, and hydrolysis is carried out by a proteolytic enzyme. |
