abstract | A novel polydeoxyribonucleic acid is disclosed having a base sequence encoding an amino acid sequence of a polypeptide which constitutes a sarcosine oxidase. This is an enzyme having the following physicochemical characteristics: it catalyzes an enzymatic reaction in which 1 mole of glycine, 1 mole of formaldehyde and 1 mole of hydrogen peroxide are obtained from 1 mole of sarcosine, 1 mole oxygen and 1 mole of water; it exhibits substrate specificity with respect to sarcosine; Optimal pH: 8.0 - 9.5; Isoelectric point: 4.7 + - 0.1; Molecular mass (determined by the gel filtration method): 40,000 + - 4,000; it is stable when treated at 40 degree (s) C for 10 minutes. Sarcosine oxidase provides a biological method for the quantitative analysis of creatinine and / or creatine, and is a major component for use not only in laboratory experiments but also in clinical diagnosis. |
