abstract | PROCEDURE AND REAGENT FOR THE DETERMINATION OF LIPASE IN BIOLOGICAL LIQUIDS. IN THIS PROCESS, AN S-ACYL COMPOUND IS USED AS THE SUBSTRATE AND A CHROMOGENIC SULPHYDRYL REAGENT TO DEVELOP MEASURABLE COLOR. THE REAGENT ALSO CONSISTS OF A SOLID AND WATER-SOLUBLE INHIBITOR OF PSEUDOCHOLINESTERASE, PREFERRED ESERINE SALICYLATE; ALBUMIN CAN BE ADDED TO INCREASE SENSITIVITY. THE LIPHASE IS DOSE BY FIRST INCUBATING THE SAMPLE TO BE DOSE WITH THE S-ACYL COMPOUND, THE CHROMOGENOUS SULFHYDRYL REAGENT AND THE INHIBITOR IN QUESTION; BY STOPPING INCUBATION USING A NON-IONIC SURFACTANT AND DIRECTLY MEASURING THE RESULTING COLOR, AND COMPARING THIS COLOR TO THAT OBTAINED WITH A STANDARD CHEMICAL SOLUTION CONTAINING S- (2-AMINOALKYL) SALT ISOTHIO-URONIUM. USE OF THESE REAGENTS AND OF THE PROCESS FOR DETERMINING LIPASE IN BIOLOGICAL FLUIDS. |
