abstract Method of measuring isoenzymes com rises placing enzymes on a support, separating the isoenzyme components contracting the support with a substrate medium, separating the substrate medium from the support, contacting the substrate medium with a precipitant, and detecting fluorescent products by fluorometry. The precipitant is (a) a mixt of 90-100 vol. % of a 1-6C alcohol and 0-10 vol. % urea (b) a 3-8C ketone, and/or (c) a soln. contg. 90-99.99% of H2O or an inert polar solvent and 0.01-10% of a salt of formula: R(Y)2 (where R is Pb2+, Ca2+, Sr2+ or Ba2+ and Y is Cl-, NO3- or ClO4-). The method is esp. useful for determining creatinine phosphokinase (CPK) and lactate dehydrogenase (LDH) isoenzymes for diagnostic purposes. The precipitant localises the fluorescent product in the substrate medium and thereby increases the sensitivity of the fluorometric measurements.
