abstract | Prepn of fluorometholone (6alpha-methyl-9alpha-fluro-11beta, 17alpha-dihydroxypregna-1,4-diene-3,20-dione) by treating 9alpha-fluoro-11beta, 17alpha-dihydroxyprogesterone with methyl or ethyl orthoformate in presence of an acid catalyst to give the 3-alkoxy-3,5-diene which is treated with a Vilsmeier reagent then rapidly hydrolysed to give the 6-formly deriv. This is a catalytically hydrolysed in presence of cyclohexene in ethanol/acetic acid to give 6alpha-methyl-9alpha-fluoro-11beta, 17alpha-dihydroxyprogesterone, which is dehydrogenated chemically or biologically to give fluorometholone. The product is an antiinflammatory steroid used in dermatology and ophthalmology. |
