abstract Improved method for the identification of microorganisms, wherein fluorescent labelled antibodies, are mixed before use, with a suitable free dyestuff, of a colour chosen either to contrast the background with the fluorescence, ( a counters train) or to neutralise the background antifluorescence, so as to provide a dark background (a "quenching" strain). Preferred dyes are acidic, and soluble in phosphate or glycine-buffered saline. The labelled antibody may be either anti-type or anti-species, should be free of foreign proteins, and pref. contain a preservative, e.g. thiomersal (0.1-10 parts/10,000), and can be a freeze-dried powder containing e.g. 75-98% gamma-globulin and up to 15% of IGA and IGM. The preferred labelling agents are fluorescein or rhodamine - B isothiocyanates and lissamine B rhodamine B, and preferred dyes rhodamine B, Evans blue etc. as counter stains, and naphthalene-black as a quenching stain.
