titleOfInvention PROCEDURE FOR OBTAINING A DERIVATIVE OF HALOGENOCOLQUICINE.
abstract Novel compounds (I): wherein R1 is a mono-, di- or tri-haloacyl radical containing 2 to 10 carbons and R2 is MeO, MeS, MeSO, MeSO 2 or NR3R4 (R3 is alkyl and R4 is H or alkyl), are obtained: (a) by acylating corresponding compounds in which R1 is H (b) by introducing a 2-methyl radical and/or an R2- substituent into a corresponding compound not containing said radical and/or substituent, and/ or into the corresponding isocolchicine compound, e.g. methylation of corresponding 2- and/or 4-hydroxy compounds (c) by reacting methyl mercaptan with a corresponding compound wherein R2 is MeO or OH, or with the corresponding isocolchicine compound, and, if desired, oxidizing the thioether (I) so obtained (d) by reacting an amine, NHR3R4, with a corresponding compound in which R2 is MeO or OH, or with the corresponding isocolchicine compound and (e) by replacing halogen in the substituent R1 by another halogen. Desacetyl - N - chloroacetyl - colchiceine is prepared by the interaction of desacetylcolchiceine and chloracetyl chloride the N- bromoacetyl compound is prepared utilizing bromoacetyl chloride. Desacetyl 14 - desmethoxy -14 - dimethylaminocolchicine is prepared by the interaction of desacetyl-colchicine and dimethylamine. Pharmaceutical preparations showing antimitotic, cytostatic and/or gout healing action contain as active ingredient compounds (I) administration is parenterally, enterally or topically.
