abstract A composition for testing whole blood for haemoglobin S comprises a buffer solution, for example a phosphate buffer pH 6.5 to 6.8, a reducing agent and an erythrocyte haemolyzing agent. The reducing agent may be sodium hydrosulphite and the haemolyzing agent a 2% solution of saponin. If the blood sample contains haemoglobin S this precipitates when the sample is mixed with the test solution, making the solution turbid.
