abstract Novel 1-acyl-3-indolyl aliphatic acid derivatives of the general formula wherein R1 is an unsubstituted or a halogeno-, C 1-2 alkyl- or phenyl-substituted or benzene ring-condensed, saturated or unsaturated mono- or poly-alicyclic group, or a C 1-4 alkyl group substituted by said alicyclic group R2 and R3 are independently a hydrogen atom or a C 1-2 alkyl group R4 is a hydrogen atom or a C 1-5 alkoxycarbonyl group R5 is a hydroxy, C 1-3 alkoxy, benzyloxy, tetrahydropyranyloxy or amino group R6 is a C 1-3 alkyl, C 1-3 alkoxy or C 1-3 alkylthio group or a halogen or hydrogen atom p is 0 or 1 and n is 0, 1, 2 or 3, are prepared by the following methods: (a) reaction of a phenylhydrazine derivative of the general formula or a salt thereof with a ketone of the general formula (b) reaction of an N1-acylated phenylhydrazone derivative of the general formula wherein B is an aldehyde or ketone residue, with a ketone of the third general formula above (c) when p is O and R5 is a hydroxy group, hydrolysis, catalytic reduction or pyrolysis of a 1-acyl-3-indolyl aliphatic ester or amide of the general formula wherein R7 is an alkoxy, benzyloxy, tetrahydropyranyloxy or amino group and (d) when p is O and R5 is an alkoxy, benzyloxy or tetrahydropyranyloxy group, reaction of a 3-indolyl aliphatic ester of the general formula wherein R7 is an alkoxy, benzyloxy or tetrahydropyranyloxy group, with an acyl compound of the general formula R1COY, wherein Y is a halogen or an ester residue. Pharmaceutical compositions having antiinflammatory, anti-pyretic, analgesic and anticholesterolemic activities comprise as active ingredient a 1-acyl-3-indolyl aliphatic acid derivative of the first general formula above and an inert, non-toxic pharmaceutical carrier or diluent. Phenylhydrazine derivatives of the second general formula above are prepared by decomposition of an N1-acylated phenyl hydrazone derivative of the fourth general formula above with an acid. N1-Acylated phenylhydrazone derivatives of the fourth general formula above are prepared by reaction of a phenylhydrazone derivative of the general formula with an acyl compound of the general formula R1COY.
