abstract Pharmaceutical compositions for oral administration comprising discrete units of pharmacologically active material are provided with at least one coating which releases the active material in the digestive tract, the coating comprising a copolymer of at least one ethylenically unsaturated monomer capable of forming a water-insoluble homopolymer, at least one quaternary ammonium compound and, if desired, at least one further copolymerisable monomer, the monomers being selected to provide a copolymer having solubility and permeability characteristics which are substantially independent of physiological pH values. Suitable unsaturated monomers include the esters of acrylic and methacrylic acid, styrene and its homologues, vinyl esters and vinyl chloride. Suitable quaternary ammonium compounds include quaternised aminoalkyl esters or aminoalkyl amides of acrylic or methacrylic acid e.g. #-methacryloyloxyethyltrimethylammonium methosulphate, #-acryloyloxypropyltrimethylammonium chloride or trimethylamino-methylmethacrylamide methosulphate. The quaternary nitrogen atom may also be a member of a heterocyclic ring, e.g. methacryloyloxyethylmethyl morpholinium chloride or the corresponding pyridinium salt, or it may be bonded to the acrylic or methacrylic group by means of a group interrupted by a hetero atom e.g. a polyglycol ether group. Quaternised vinyl-substituted nitrogen-containing heterocyclic compounds such as methylvinyl pyridinium salts, vinyl esters of quaternised aminocarboxylic acids and styryl trialkylammonium salts may also be used The solubility of the coating may be modified by the addition of suitable monomers such as acrylamide, methacrylamide, hydroxy-alkyl esters of acrylic or methacrylic acid and vinyl pyrrolidone.
