abstract | Method for qualitatively and quantitatively determining the presence of an anticoagulant in a sample using prothrombinase-induced clotting time (PiCT), wherein the presence of anticoagulants in the sample prolongs, in a dose-dependent manner, the time to clotting, wherein said method comprises: (1) the use of standard PiCT, which comprises adding a defined amount of PiCT activator to the sample, incubating at 37°C for a specific period of time, adding the starting reagent and measuring the time to coagulation in said reaction mixture; (2) use of an adapted version such as inverted PiCT, comprising adding the starting reagent to the sample, incubating at 37°C for a specified period of time, adding a defined amount of PiCT activator, and measuring the time until coagulating said reaction mixture; (3) the use of reference intervals that are tailored to the detection system and assay method, where the reference intervals are compared to clotting times in the reaction mixture and where reaction times outside the range in the reaction mixture indicate the presence of anticoagulants; and (4) confirmation of clotting times in the reaction mixture using activated partial thrombin time (aPTT) and prothrombin time (PT) to identify the nature of the anticoagulant; wherein the sample is selected from blood or plasma, and the anticoagulants are selected from direct or indirect FIIa or FXa anticoagulants. |
