A glucoamylase variant, comprising a substitution at position T43, wherein the position corresponds to amino acid positions in the amino acid sequence set forth in SEQ ID NO: 3; and wherein the variant comprises at least one of the following combinations of substitutions: T43K+S95P+A121P+Y295W+Q318Y; V18M+T43K+S95P+A121P+Y295W+Q318Y; D4R+T43K+S95P+A121P+Y295W+Q318Y; S5V+T43K+S95P+A121P+Y295W+Q318Y; I13S+T43K+S95P+A121P+Y295W+Q318Y; wherein the variant has glucoamylase activity and wherein the variant has at least 75%, at least 80%, at least 85%, at least 90%, at least 95%, at least 96% , at least 97%, at least 98%, or at least 99%, but less than 100% sequence identity to the polypeptide of SEQ ID NO: 3, and variants have increased melting temperature as measured by TSA of at least 2 °C, particularly at least 3 °C compared to the glucoamylase of SEQ ID NO: 3. |
