abstract A method for the simultaneous detection of multiple analytes in a sample, comprising: (a) contacting the sample with a set of binding agents conjugated to a plurality of isotope-encoded indicators (iCORE), wherein the binding agents are capable of specifically binding a set of analytes, under conditions suitable for the binding agents to bind to the analytes, wherein the set of binding agents comprises at least 5, at least 6, at least 7, at least 8 , at least 9, at least 10, at least 11, at least 12, at least 13, at least 14, at least 15, at least 16, at least 17, at least 18, at least 19 or at least 20 agents binding conjugates to different iCOREs, wherein the iCOREs are peptide mass tags and wherein (i) the sample is contacted in vivo or (ii) the binding agents are conjugated to the iCOREs via a photocleavable linker; (b) identifying and/or quantifying, optionally by MS/MS assay, the iCOREs of the binding agents bound to the analyte; and (c) identifying and/or quantifying the analytes present in the sample based on the iCOREs identified in (b).
