titleOfInvention | Methods and compositions for rapid plant transformation |
abstract | A method of producing a transgenic plant, comprising: (A) (a) transforming a cell from an immature embryonic explant with an expression construct comprising: (i) a nucleotide sequence encoding a WUS/WOX homeo sequence polypeptide ; (ii) a nucleotide sequence encoding a polypeptide comprising two AP2-DNA binding domains; or (iii) a combination of (i) and (ii); (b) allowing expression of the polypeptide from (a) in each transformed cell to form a regenerable plant structure in the absence of exogenous cytokinin, in which no callus is formed and the regenerable plant structure is formed within 1-7 days of transformation of the cell or in 1-14 days of cell transformation; and (c) germinating the regenerable plant structure to form the transgenic plant; or (B) (a) transforming a cell from an immature embryonic explant with an expression construct comprising: (i) a nucleotide sequence encoding a WUS2 polypeptide; (ii) a nucleotide sequence encoding an ODP2 polypeptide; or (iii) a combination thereof; (b) allowing expression of a polypeptide of (a) in each transformed cell to form a regenerable plant structure in the absence of exogenous cytokinin, in which callus is not formed; and (c) germinating the regenerable plant structure to form the transgenic plant; wherein the WUS2 polypeptide comprises the amino acid sequence of SEQ ID NO: 4; or wherein the WUS2 polypeptide is encoded by the nucleotide sequence of SEQ ID NO: 3; and wherein the ODP2 polypeptide comprises the amino acid sequence of SEQ ID NO: 18; or wherein the polypeptide comprising two AP2-DNA binding domains is encoded by the nucleotide sequence of any one of SEQ ID NO: 17 or 21. |
