abstract A method of orienting, processing, embedding and microtome cutting a tissue sample (16) using a block of gel compound (12) preformed into a self-supporting geometric shape, the method comprising: retaining the tissue sample (16) in an orientation in a hole, indentation, or slot formed in the geometrically shaped, preformed gel compound block (12), and between first and second resiliently deformable portions (20, 22) of the same shaped, preformed gel compound block geometry (12), the first and second resiliently deformable portions applying a force to the tissue sample (16) to thereby retain the tissue sample (16); processing the tissue sample (16) while it is oriented, subjecting the tissue sample (16) and the geometrically shaped, preformed gel compound block (12) to processing fluids and reagents, fixing the preformed gel compound block, and geometrically shaped (12) and retained tissue sample (16) in a microtome sectionable cassette (30) embed the preformed geometrically shaped gel compound block (12), the microtome sectionable cassette (30) and the tissue sample while oriented in an embedding medium (38) to form a microtome sectionable block of the embedding medium, the tissue sample, the microtome sectionable cassette (30), and the preformed and shaped gel compound block geometric (12); and microtome sectioning the sectionable block to obtain thin sections of the diagnostic tissue sample.
