A method of isolating a lipid containing polyunsaturated fatty acids (PUFA) from a biomass, comprising the following steps: a) providing a suspension of a biomass comprising cells containing a lipid containing PUFA; b) optionally lysing the cells of the biomass; c) concentrating the suspension to a total dry matter (TDM) content of 20 to 60% by weight, if the suspension has a lower TDM content; d) adjusting in the suspension a temperature of from 20°C to 100°C, preferably from 25, 30, 40, 50 or 60°C to 100°C, more preferably from 65°C to 95°C, in particular from 70 to 90°C; e) maintaining the temperature in the range described in (d) for at least 1 hour, preferably for at least 2, 3, 4, 5, 6, 7 or 8 hours, more preferably for 1 to 36 hours, in particular during from 2, 3, 4, 5, 6, 7 or 8 hours to 36 hours, more preferably from 10 to 24 hours, while adding a total of 7.5 to 25 moles, preferably 8.5 to 22 moles , in particular from 10 to 20, more preferably from 11 to 18, especially from 12 to 17 moles, of base equivalent to 10 kg of total dry matter contained in the suspension. |
