abstract | A method of identifying the fertility status of a human male subject comprising: exposing a first portion of a sperm sample from the subject to non-capacitating conditions to obtain non-capacitating sperm; exposing a second portion of the sperm sample to capacitating conditions to obtain capacitated sperm; fix the capacitated sperm and the non-capacitated sperm with a fixative; stain capacitated sperm and non-capacitated sperm to identify GM1 patterns; determining a frequency of selected GM1 patterns in capacitated sperm and non-capacitated sperm; and comparing the frequency of selected GM1 patterns in capacitated sperm with the frequency of selected GM1 patterns in uncapacitated sperm, wherein a change in the frequency of one or more of the selected GM1 patterns in sperm capacitated from uncapacitated sperm is indicative of the fertility status of the individual, in which the GM1 patterns selected are one or more of intermediate (INTER), apical acrosome (AA) and post-acrosomal plasma membrane (APM). |
