abstract | A method of identifying a microorganism in a liquid comprising interfering proteins from a mammalian source, wherein the interfering proteins preferably include one or more hemoglobin, defensins or their proteolysis products, the method comprises: preparing a lysate, the lysate comprises the interfering proteins and the proteins of the microorganism; contacting the lysate with a chromatographic medium, where the interfering proteins and the proteins of the microorganism bind to the chromatographic medium; selectively eluting the bound proteins to the chromatographic medium to produce at least one eluted fraction, wherein the at least one eluted fraction is enriched in microorganism proteins and reduced in interfering proteins; and subjecting the at least one eluted fraction to protein mass spectrometry analysis, such as MALDI or ESI-MS, to identify the presence of one or more microorganisms in the liquid where the liquid is preferably a blood one, a blood culture, urine or cerebrospinal fluid. |
