abstract Method for the absolute quantification of a peptide ligand of the MHC located on a cell, in which said method comprises a) the preparation of the cells that present said peptide ligand of the MHC from a biological sample that is selected from a tissue sample, a blood sample, a tumor sample or an infected tissue sample, comprising said cells, comprising enzymatic digestion of tissues, and/or cell lysis b) determination of the number of cells of said preparation of step a) , which includes counting cell nuclei, DNA photometry, DNA fluorometry, or quantitative PCR, c) addition of a known amount of the peptide ligand of the MHC and/or the peptide ligand-MHC complex that is intended to be quantified resulting from said preparation of step a) directly after tissue homogenization ("1st enrichment"), d) isolation of the MHC peptide ligand resulting from said preparation ion of step c) in order to obtain a peptide eluate, e) addition of a known amount of the MHC peptide ligand to be quantified to said peptide eluate ("2nd enrichment"), as an internal calibrator, f) execution of a mass spectrometry analysis of said peptide ligand of the MHC in order to generate: aa) a signal to determine the efficiency of the isolation carried out in step d), bb) a signal of the internal calibrator of step e ), and cc) a signal corresponding to said MHC peptide ligand from the cells prepared in step a), and g) quantification of said MHC peptide ligand based on the comparison of the signals obtained in step f) with: aa ) the cell count obtained, bb) the known amount of said MHC peptide ligand and/or peptide ligand-MHC complex that is to be quantified and that has been added in step c), and cc) the known amount of the ligand MHC peptide that is intended to quantify and that has been added in step e), in which the quantification comprises the calculation of a ratio between the signal of the internal calibrator of step e) and the signal of the isolated MHC peptide ligand, and the comparison of the ratio with an established calibration curve of the MHC peptide ligand, and wherein the quantification further comprises generating a peptide-specific calibration curve based on a ratio obtained with the internal calibrator used in the same amount, and determining the lower limit of quantification (LIC) for said peptide ligand of the MHC that is intended to be quantified, with which the absolute quantification of the peptide ligand of the MHC shown by the cell is achieved in the case of the peptide ligand of the MHC of interest if the quantified amount exceeds the determined LIC.
