abstract | A method for removing contaminating DNA in a sample containing antibodies comprising the following steps: 1) applying the sample containing the antibody to Protein A affinity chromatography, followed by an elution step involving i) equilibration of the Protein A resin using 1 M NaCl / 10 to 100 mM citrate-phosphate buffer at pH 7.5; ii) loading the antibody sample onto the resin, followed by iii) a first washing step using 1 M NaCl / 10 to 100 mM citrate-phosphate buffer pH 7.5, and iv) a second washing step using 10 to 100 mM citrate-phosphate buffer pH 7.5, followed by v) an elution step using 2.5 mM HCl pH 2.6; 2) adjust the pH of the eluate to pH 7 with a 300 mM Tris solution to form particles; and 3) removing the resulting particles, wherein the antibody is the humanized anti-IL-6 receptor hPM-1 antibody. |
