abstract An isolated cell comprising a cellular biosensor system comprising at least one gene (1) (REPRESSOR MODULE IA) comprising 1.1 a promoter sequence (P1), 1.2 a transcriptional activator sequence (TA) operably linked to the promoter sequence (P1 ), 1.3 at least one target site (T1) for one or more entries (IN), in which, upon binding of the entry (IN), the target site (T1) inhibits or represses the expression of the transcriptional activating sequence (TA) operably linked to the promoter sequence (P1); at least one gene (2) (REPRESSOR MODULE IB) comprising. 2.1 an activator-inducible promoter (P2) expressed by the transcriptional activating sequence (TA) and operably linked to, (i) a transcriptional repressor sequence (TR) and at least one target site (T2) for one or more inputs (IN ), and / or (ii) one or more repressor sequences encoding at least one repressor microRNA (R-miRNA), in which, upon entry binding (IN), the target site (T2) inhibits or represses the expression of the transcriptional repressor sequence (TR), and wherein the expressed transcriptional repressor (TR) and / or the at least one repressor microRNA (RmiRNA) inhibit or repress the expression of at least one exit sequence from an exit module (3); at least one gene (3) (OUTPUT MODULE) 3.1 comprising a promoter sequence (P3) that is repressible by the product expressed by the transcriptional repressor sequence (TR) of gene (2), if gene (2) comprises the transcriptional repressor sequence (TR), 3.2 at least one exit sequence, 3.3 at least one sequence or sequences that allow gene rearrangement (SEGR) in the presence of at least one site-specific recombinase (Rec1), 3.4 at least one target site (TmiRNA) for the at least one repressor microRNA (R-miRNA), if the gene (2) comprises one or more repressor sequences encoding at least one repressor microRNA (R-miRNA), wherein, after gene rearrangement in presence of at least one site-specific recombinase (Rec1), the promoter (P3) is operably linked to the exit sequence 3.2 to generate the exit sequence; and at least one gene (4) (RECOMBINASE MODULE) comprising 4.1 at least one promoter (P4), 4.2 a sequence encoding the site-specific recombinase (Rec1) that allows gene rearrangement in gene (3) and is operably linked to the promoter (P4), 4.3 optionally at least one target site (T3) for one or more entries to inhibit or repress the expression of site-specific recombinase (Rec1), and 4.4 optionally at least one target site (TmiRNA) for the at least one repressor microRNA (RmiRNA), if the gene (2) comprises one or more repressor sequences encoding at least one repressor microRNA (RmiRNA).
